چکیده

Michael J. Antonaccio and Benedict R. Lucchesi Department of Pharmacology, The University of Michigan Medical School, Ann Arbor, Michigan 48104 (Received 22 May 1970 ; in final form 3 August 1970) Glucagon has been shown to be both a positive inotropic and chronotropic agent in many species including man (1-5). However, the positive inotropic effect of glucagon on dog papillary muscles can be reversed by previous exposure of the tissues to a relatively high concentration of ouabain (2). The purpose of this study was to determine the interactions of glucagon on dog isolated papillary muscles with other positive inotropic agents which presumably have a different mechanism of action .

Cardiac actions of glucagon.

Glucagon increased heart rate and contractile force in the anesthetized dog and increased isometric tension in the isolated dog papillary muscle. The positive inotropic and chronotropic responses were not abolished or reduced by previous administration of the beta-receptor blocking agent propranolol, in doses of 0.5 mg/kg. Glucagon increased the force and rate of cardiac contraction even when c...

Correlation of cyclic AMP and cyclic GMP levels with changes in contractile force of dog ventricular myocardium during cholinergic antagonism of positive inotropic actions of histamine, glucagon, theophylline and papaverine.

On the dog isolated right ventricular muscle, experiments were carried out in order to elucidate the characteristics of the cholinergic antagonism against the positive inotropic action (PIA) induced via different subcellular mechanisms. The relationship between cyclic AMP and cyclic GMP levels, and contractile force during cholinergic antagonism was assessed. Carbachol (10 microM) by itself dec...

Glucagon. Its enhancement of cardiac performance in the cat and dog and persistence of its inotropic action despite beta-receptor blockade with propranolol.

The action of glucagon on cardiac performance was studied -in 21 isolated cat papillary muscle preparations, in 13 spontaneously beating cat atria, in 15 intact dog hearts, and in 4 isolated perfused dog hindlimbs. In each papillary muscle preparation, addition of glucagon produced marked increases in maximal developed tension, averaging 36 ± 4.2% (SEM) (P<0.01) , and shifted the force-velocity...

Effect of Sodium Valproate on Ouabain-Induced Arrhythmia in Isolated Guinea-Pig Atria

Sodium valproate (SV), an antiepileptic drug has several mechanism of action. It inhibits voltage sensitive Na+ channels and reduces intracellular Na accumulation. These actions are similar to that of both phenytoin and carbamazepine. We have investigated the direct cardiac action of SV and its effects on ouabain-induced arrhythmia in isolated guinea-pig atria. The guinea-pig atrium was dissect...

Enhancement of sodium-calcium exchange induces positive inotropic action and potentiates ouabain effect in rat hearts.

To study the inotropic effect of enhanced Na(+)-Ca(2+) exchange in the rat papillary muscles and isolated heart, the developed tension in the rat papillary muscles was measured and the left ventricular functions were assessed in the isolated rat heart. E-4031, a selective activator for Na(+)-Ca(2+) exchange in rats, concentration-dependently increased the developed contractile tension in the ra...
